After a round consisting of seven birdies and an eagle, Perry sits on 65 to be five strokes clear of the field.
Preparation has been key for Perry who credits his early lead to recent practice at Commonwealth.
“I had the opportunity to play the course the other day and it really paid off, I hit a lot of greens in regulation,” said Perry.
Peter Wilson sits in second place on 3-under 70, one shot clear of Charlie Dann on 2-under par with two rounds still to play.
Commonwealth Golf Club was in excellent condition with firm and fast greens greeting players, as well as sunny skies and light winds.
The Melbourne Challenge golf action moves to Kingston Heath Golf Club on 22 January and Sandhurst Golf Club on 23 January.
